Incorporarse al laboratorio del grupo de Lipotoxicidad y enfermedad del hígado grado bajo la dirección del Profesor Antonio Vidal-Puig en colaboración con la Dra. Stefania Carobbio en el Centro de Investigación Príncipe Felipe. El/la candidata/a contribuirá a nuestra investigación que se centra en refinar la identificación de factores intrahepáticos regulados tempranamente que influyen en la transición de la esteatohepatitis metabólica asociada al metabolismo (MASLD) a la esteatohepatitis asociada a disfunción metabólica (MASH) y evaluar su potencial como dianas terapéuticas y biomarcadores diagnósticos/pronósticos.

Los diferentes objetivos del proyecto incluyen trabajos tanto in vivo como in vitro en forma de a) Estudios longitudinales relacionados con la exposición en ratones (varias dietas lipídicas, temperatura ambiental y GLP1) para definir las trayectorias naturales y los posibles mecanismos de reversión MASLD a MASH b) Validación in vitro: examinar la regulación de las redes moleculares bajo estrés genético y ambiental utilizando modelos celulares hepáticos c) Validación in vivo: confirmar la relevancia clínica de los genes y vías identificados en la progresión de MASLD a MASH mediante estudios in vivo murinos y humanos.

El trabajo del titular del puesto implicará múltiples responsabilidades básicas en un entorno de laboratorio, incluido el procesamiento y la gestión de muestras tomadas como parte de estudios en curso, cultivo celular, aplicación rutinaria de técnicas estándar de biología celular y molecular como extracción de lípidos/ARN/proteínas, genotipado, RT-PCR, western blotting, citometría de flujo y microscopía confocal. Además, el candidato seleccionado colaborará en el fenotipado de animales y la recogida de tejidos. El titular del puesto también deberá gestionar la introducción o el análisis de datos, será responsable del pedido de consumibles y contribuirá al mantenimiento del equipo central. El candidato seleccionado deberá estar muy motivado, ser un comunicador eficaz con buenas aptitudes para la informática, la gestión de datos y la organización, y tener capacidad para trabajar de forma independiente o en equipo, según sea necesario.

Join the Lipotoxicity and Fatty liver disease laboratory under the direction of Professor Antonio Vidal-Puig's (I-81) in collaboration with Dr. Stefania Carobbio at Centro de Investigacion Principe Felipe. The candidate will contribute to our research that focuses on refining the identification of early regulated intrahepatic factors that influence the transition from Metabolic metabolic-associated steatohepatitis (MASLD) to Metabolic Disfunction-Associated Steatohepatitis (MASH) and evaluate their potential as therapeutic targets and diagnostic/prognostic biomarkers.

The different objectives of the project include both in vivo and in vitro work in the form of a) Longitudinal exposome-related studies in mice (several lipid diets, environmental temperature and GLP1) to define the natural trajectories and potential reversal mechanisms MASLD to MASH b) Validation in vitro: to examine the regulation of molecular networks under genetic and environmental stresses using hepatic cellular models c) In Vivo Validation: to confirm the clinical relevance of identified genes and pathways in the progression of MASLD to MASH through murine and human in vivo studies.

The post holder's work will involve multiple core responsibilities within a laboratory environment, including the processing and management of samples taken as part of ongoing studies, cell culture, routine application of standard cell and molecular biology techniques such as lipid/RNA/protein extraction, genotyping, RT-PCR, western blotting, flow cytometry, and confocal microscopy. Additionally, the successful applicant will assist with animal phenotyping and tissue collection. The post holder will also be required to manage data entry or analysis, will be responsible for ordering consumables, and will contribute to the maintenance of core equipment. The successful candidate will be a highly motivated, effective communicator with good IT, data management, and organizational skills and the ability to work independently or within a team as required.
